Sažetak
Dendritic cells (DCs) represent a heterogeneous cell population that balances immune response towards immunity or tolerance. Upon pathogen capture, DCs migrate to the secondary lymphoid organs and activate naive T cells to initiate adaptive immune responses. Their role in pregnancy is still unclear. We analyzed presence, distribution, origin and function of DCs in normal human first trimester decidua and DC subsets in mice. Experiments were performed on freshly isolated and/or cultured decidual mononuclear cells (DMC) of normal human first trimester decidual tissue and adult C57Bl/6 female mice mated with syngenic (C57Bl/6) and allogenic (BALB/c) males. DCs were detected by flow cytometry and immunohistochemistry. Human CD83+ cells were purified and used in CFSE proliferation assay. Decidual CD83+ cells up-regulate HLA-DR and CD86, while they down-regulate mannose receptor (CD206) after over night culture. CD83+ cells in the suspension of DMCs efficiently took up antigen. After 72 hours cultivation with purified CD83+ cells, an increased proliferation rate of decidual CD56+ cells was obtained. In both syngenic and allogenic murin pregnancy model (7 day of gestation), number of uterine CD11c+ cells was increased and the expression of CD86 and MHC class II (I-Ab) molecules on their surface was up-regulated, compared to the virgin ones. At the materno-fetal interface, human (CD11c+highCD123low) and murin (CD11c+CD8-) DCs are of myeloid origin. Decidual DCs express high amount of co-stimulatory and MHC class II molecules and are capable of activating decidual NK cells.
