Sažetak
Genetic changes of two tumor suppressor genes: Adenomatous polyposis coli (APC) and E-cadherin (CDH1) were investigated in squamous cell carcinoma of the larinx. APC gene instability was tested by polymerase chain reaction/loss of heterozygosity (LOH) using Restriction Fragment Length Polymorphism (RFLP) method. The results of our analysis showed 3 samples with LOH of the APC gene out of 15 heterozygous patients (20%) when tumor DNA was compared to autologous constitutive DNA. When we detected changes of the APC gene in our sample, we went on to E-cadherin suppressor gene analysis. The gene is connected to APC both in adherens junctions, and in the wnt signal transduction pathway. CDH1 genetic changes were analyzed using PCR amplification of D16S752 marker (informativeness 91%, Pećina-Šlaus N et al., Coll Antropol, 2002, 26 ; 85-8) linked to CDH1 gene. By Spreadex gel electrophoresis (Elchrom Scientific, Switzerland) one LOH of CDH1 gene (5, 55%) out of 18 heterozygous patients was discovered. Replication error positive samples were not detected in our sample. Our results suggest that alterations, in APC gene may be responsible for squamous cell carcinoma development and progression. The results were obtained by usage of limited number of genetic markers and we propose additional work for future research.
