Sažetak
Cell populations in the decidua of pregnancy are heterogeneous and besides stromal cells include bone marrow derived immune cells: macrophages, large granular lymphocytes (LGL) and T lymphocytes. Immune cells infiltrating decidua are constantly targeted by placental hormones and cytokines, produced at the materno-fetal interface and acting in a paracrine or autocrine manner. Cytokines of TH2 type (IL-4, IL-6, IL-10, IL-15, TGFbeta) seem to be predominantly produced rather than those of TH1 type. LGL have an NK like phenotype (CD3+CD56brightCD16+), they are armed with the strong cytolytic potential (Perforin-Pbright+), but exhibiting much lower cytotoxic potential than corresponding PBL. Experiments with perforin deficient animals showed the role of P+ cells in homeostatic regulation of the immune response by lyzing antigen presenting cells and preventing excessive T cell activation. From the other side, decidual macrophages have an important role in maintenance and up-regulation of P expression in decidual LGL. These effects are mediated by humoral products and could be ascribed to IL-15 and progesterone, and its mediator (PIBF). IL-2, the most potent stimulator of P expression in PBL, is almost completely absent from the decidua, as well as his high affinity receptor (CD25), which can explain IL-2 inefficiency in upregulation of P expression. In decidua IL-15 exhibits many biological activities ascribed to IL-2, including upregulation of P expression. The role of cytokines IL-10 and IL-12 and IFNgama was also investigated. The results point to the existence of hormonal/cytokine network regulating activation of cytolytic pathways at the maternal-fetal interface.
