Sažetak
Feto-placental unit in mammals is in a direct and intimate contact with the decidua infiltrating maternal immunocompetent cells with a potent cytolytic machinery. The prevalent phenotype of these cells is CD3-CD56bright+CD16- but full of perforin (P ; Pbright+). It is well known that several cytokines can induce and upregulate the expression of P in the peripheral blood lymphocytes (IL-2, IL-6, IL-7, IL-12), IL-2 being the most potent. However, there is an almost complete absence of IL-2 and its high affinity receptor (IL-2R ; CD25 molecule) at the materno-fetal interface. IL-15 has been found to share many immunoregulatory activities with IL-2, and IL-15 mRNA has been detected in a number of human tissues including placenta. We hypothesized that IL-15 would therefore be an important factor regulating perforin expression in decidual lymphocytes. Our results with first trimester pregnancy decidual lymphocytes clearly showed that P expression in cultured decidual non adherent cells (DNAC) and peripheral blood lymphocytes (PBL) can be upregulated by coculture with decidual adherent cells (DAC), or their supernatants, in 18h culture. Immunocytochemistry staining has shown that 80% of DAC represent stromal cells and 10-20 % macrophages. Immunohistology staining of serial frozen first trimester decidual tissue sections showed the same distribution of macrophages (Ber-Mac-3+ cells) as IL-15+ cells, indicating that cells responsible for secreting IL-15 could be predominantly macrophages. Effects of DAC on P expression in DNAC can be blocked completely by addition of monoclonal anti-IL-15 antibodies. Contrary, P expression in peripheral blood non adherent cells (PBNAC) mediated by DAC could be blocked only partially by addition of anti-IL-15. Upregulation of P expression was obtained by stimulation of DNAC with IL-15, but not with IL-2.
