Sažetak
Large granular lymphocytes of NK like phenotype (CD3-CD56+brightCD16-), which contain high level of cytolytic protein perforin (P) in their granules (bright+) are prevalent bone marrow derived cell population in the pregnancy decidua. These cells make 85-90% of non-adherent cell fraction. The number of P+ cells significantly drop after 18 hours incubation in vitro, but can be completely prevented by addition of decidual and peripheral blood adherent cells and their supernatants. Therefore, we investigated phenotype of decidual adherent cells (DAC) and peripheral blood adherent cells (PBAC) in the first trimester pregnancy. Peripheral blood adherent cells (PBAC) are homogenous population of monocytes, while DAC are heterogeneous cell populations that prevalently do not express monocyte/macrophage markers (cca 20% CD45 ; 5-15% CD14, HLA-DR and CD11c). By immunocytochemistry we found that about 80% of DAC are at the same time vimentin+, citokeratin-, -actin+ and epidermal growth factor receptor (EGFR) positive, indicating that they are decidual stromal cells. Macrophages make further 10-20% and only 5% of the cells belong to glandular epitel (citokeratin+) and endothel of spiral artery (Q-band 40+). To clarify which cell populations affect P expression, we compared the effects of DAC, and HLA-DR depleted DAC. Contrary to DAC, the HLA-DR depleted cells (mostly stromal cells) can not prevent the lost of P in DNAC, and in PBNAC. Macrophages, as issue of many cytokines, presented only in DAC, seem to be strong regulators of perforin expression in decidua. Antibodies anti IL 10, but not IL 12, were efficient in preventing the effects of macrophages on maintaining the P level. The results are pointing to the importance of macrophages and Th2 type cytokines in the control of activation of cytolytic pathways in decidua.
