Sažetak
Althaeae radix (obtained from Althaea officinalis L., Malvaceae) contains mucilages (about 25-35%) with galacturonic acid, glucuronic acid and rhamnose as major components. Due to the specific composition with a lot of mucilage and starch, such herbal drugs are often a subject of microbiological contamination. Although γ -irradiation is a contemporary effective, economic, fast, practical and commercial method for a microbial decontamination, it cannot be applied when herbal drugs are abundant with mucilage, since these substances will be degraded. Our study aimed to find out what happened to the free amino acids when this drug was exposed to γ -irradiation doses of 1, 5 and 10 kGy. These doses are most frequently used for a microbial decontamination. By a TLC method it has been discovered that Althaeae radix contained free amino acids as follows: γ -aminobutyric acid (Gaba), alanine (Ala), arginine (Arg), asparagine (Asn), lysine (Lys), as well as some other amino acids in smaller quantities. TLC method has been performed on cellulose and silica gel, using solvent system: n-butanol - acetone - glacial acetic acid - water (35:35:10:20 V/V/V/V). Detection has been carried out by a ninhydrin reagent. The TLC method proved that the tested samples of Althaeae radix contained identical amino acids pattern irrespective of the γ -irradiation treatment.
