Sažetak
In an attempt to obtain a model resembling natural listeriosis, we studied the course of Listeria monocytogenes infection in mice challanged intragastrically (i.g.). Balb/c mice received different doses of EGD strain of L. monocytogenes, directly into the stomach after overnight fasting. At different time points we followed the bacterial load, pathohistological and histochemical findings in different organs. In order to evaluate the contribution of CC-chemokines in vivo, the levels of MIP-1alpha, MCP-1 and RANTES were determined by enzyme immunoassay in murine sera and organ homogenates. Single, high chalenge doses of L. monocytogenes resulted in severe illness and a lethal outcome while repeated i.g. administration of lower doses for three consecutive days significantlly reduced the mortality rate. However, systemic infection was always achieved according to high number of listeria recovered from the liver, spleen, lungs, kidney and, suprisingly, from the brain of all infected animals. During the inflammatory event, chemokine concentrations typically increased more at the local site compared to the systemic level. The gradient between a local compartment and the intravascular space was quantified by determing the ratio between the organ and serum levels of the chemokines.The liver to serum ratio was the most pronounced in the case of MIP-1alpha, suggesting its possible role in the necroinflammatory response in the liver. According to the ratio of brain to serum MIP-1alpha and RANTES levels, in the brain tissue a reverse chemotactic gradient was present. The ratio of local to systemic chemokine concentrations, rather than absolute organ or serum values, seems to be useful in predicting the severity of inflammatory response. A greater uderstanding of the role that chemokines play may be important for explaining the immunopathogenesis of listeriosis, as well as, for planning future therapeutic protocols.
