Sažetak
Post mortem brain studies indicated that serotonin (5-hydroxytryptamine, 5-HT) could be involved in the ethiology of Alzheimer's disease (AD). The aim of the study was to determine platelet 5-HT concentration and platelet monoamine oxidase type B (MAO) activity in patients with early-onset AD and late-onset AD and in healthy subjects. The diagnosis of the probable Alzheimer's disease (NINCDS-ADRDA criteria) was established by two psychiatrists, according to ICD-10 and DSM-IV-TR criteria. The study included 52 patients with AD (23 men ; 29 women) and 75 healthy controls (36 men ; 39 women). In 25 patients (17 men, 8 women) the symptoms of early-onset AD (before the age of 65 years ; range 50-65 years) were observed. Late- onset AD was detected in 27 patients (6 men, 21 women) with age range from 67 to 88 years. Platelet 5-HT concentration and platelet MAO activity were determined spectrofluorimetrically. All participants gave their informed consent. Data were evaluated using one-way analysis of variance (ANOVA) followed by Newman-Keul's t-test. There was no sex related difference in platelet 5-HT concentration between patients with AD and healthy controls. Patients with early-onset AD had similar platelet 5-HT values as sex-matched patients with late-onset AD and healthy controls. Significant (F=7.31 ; df=3, 123 ; p<0.001, ANOVA) difference in platelet MAO activity was observed between groups, with higher platelet MAO activity in male and female patients with AD than in corresponding healthy controls (p<0.05 Newman-Keul's). A significant difference (F=5, 77 ; df=5, 121 ; p<0.001, ANOVA) in platelet MAO activity was found between patients with early- and late-onset AD and healthy controls. The highest values (p<0.05 Newman-Keul's) of platelet MAO activity were found in both male and female patients with late-onset of AD when compared to corresponding healthy controls. The results from our preliminary, ongoing study, support the presumption that platelet MAO activity could be used as a biological marker for different categories of AD.
