Sažetak
Intestinal lactase-phlorizin hydrolase, sucrase-isomaltase and maltase-glucoamylase are brush-border hydrolases important for the digestion of carbohydrates. Alkaline phosphatase is a metalloenzyme generally used as a marker of epithelial cell maturation. The expression of these enzymes during postnatal development has been characterized, but the effect of ageing on disaccharidase activity is still poorly understood. Our aim was to determine the influence of ageing on lactase, sucrase, maltase and alkaline phosphatase activities at five equidistant segments of the gastrointestinal tract (duodenum, jejunum, ileum, colon and rectum). BALB/c mices, 3-, 6- and 12-month-old were used in this study. Intestinal disaccharidase activities were determined according to the Dalquist method. Alkaline phosphatase activity in the small intestine was determined according to the method of Bowers and McComb. This study has revealed that the total intestinal disaccharidase activities in 12-month-old mice were significantly lower than those in 3-month-old animals (p<0, 05). Intestinal alkaline phosphatase activities in 12-month-old animals were significantly increased compared to 3-month-old mice (p<0, 05). The age related changes in disaccharidase activities and alkaline phosphatase activity in BALB/c mice could contribute to a reduced absorption of nutrients and consequently lead to an increased incidence of malnutrition in elderly mice. Further studies should determine if there is a connection between ageing and the brush-border enzymes activity in humans.
