Sažetak
We studied consequences of Mre11 dysfunction in an Arabidopsis line with a T-DNA insertion in the conserved N-terminal region of the AtMre11 gene. Plants homozygous for the insertion are viable but they exhibit severe growth defects. Cytogenetic analyses of mitotic sperads revealed abundant dicentric chromosomes and chromosome fragments. The frequency of chromosome fusions did not decrease in mre11 ku70 double mutants suggesting that plants possess robust DNA end-joining acticities that are independent of Ku and Mre11 complexes. The presence of subtelomeric sequences at a portion of anaphase bridges, detected by FISH, indicated that these bridges may result from chromosome fusions. The presence of end-to-end chromosome fusions in mre11 mutants was further confirmed by cloning telomere-telomere fusion junctions by PCR using primers specific for subtelomeric sequences. Interestingly, the average length of telomeres in mre11 mutants did not differ from that in the wild type control although the amount of telomeric DNA remaining at the telomere-telomere fusion junctions was substantially lower than the shortest detected telomeres. These data indicate that the genome instabilities observed in Mre11-deficient cells are, at least in part, derived from a stochastic loss of telomere function at a subset of chromosome termini.
