Sažetak
DNA vaccine consist of a bacterial plasmid with a strong viral promoter, the gene of interest and a polyadenilation/transcriptional termination sequence. The plasmid is grown in bacteria (E.coli), purified, dissolved in a saline solution, and then simply injected into the host where the encoded protein is made. DNA vaccines have been shown to induce both humoral and cellular immune responses in a variety of preclinical animal models for viral, bacterial and parasitic diseases. In this review we summarise current knowledge on the construction of DNA vaccines, immunological mechanisms, safety considerations and preclinical disease models. We discuss some of the obstacles that remain to be overcome en route to successful DNA vaccination. If proven to be safe, the DNA vaccines can serve as an alternative to classical vaccines.
