Sažetak
In the present work we investigated isoforms of stress-70 proteins in the gills of mussel M. galloprovinicialis using 2-D gel electrophoresis. 1-D analysis (SDS-PAGE) using monoclonal mouse antibodies anti-HSP70 detected two bands of stress-70 proteins 70 and 72 kDa. 2-D analysis (IEF + SDS-PAGE + WB) proved that used antibodies detected HSP70 (pI 5.7-5.9) and HSP72 (pI 5.5-5.6). Image analysis of 2-D gels stained with GelCode Blue (commasie) and corresponding Western Blot were done using gel comparison program Flicker (http://www.lecb.ncifcrf.gov/flicker/) which runs on any www connected computer and is invoked from a Java-capable browser. Results indicated that it is possible detection and quantification of stress-70 protein isoforms on 2-D stained gels without WB analysis. The usefulness and problems of such detection of stress-70 proteins as biomarkers of environmental pollution is discussed.
