Sažetak
Cancer cells show variable adenovirus vector uptake. Some studies showed the correlation between the efficacy of adenoviral infection and the expression of primary adenovirus receptor CAR (coxsackie adenovirus receptor), while other studies attributed this to expression of secondary receptors i.e. integrins. We developed cisplatin resistant CA3ST cells from cisplatin sensitive human laryngeal carcinoma cells HEp2, and found that CA3ST cells are 5-fold more sensitive to adenoviral transduction than HEp2 cells. The aim of this study was to investigate the mechanism initiated by drug resistance development, which led to the increased adenoviral transduction efficacy. RT-PCR analysis revealed up-regulation of mRNA for b3 integrin in CA3ST cells. Using flow cytometry we confirmed up-regulation of avb3 integrin in CA3ST cells and found moderately increased expression of CAR. HEp2 derived cell lines expressing different amounts of avb3 integrin have been isolated in order to test its implication in transduction efficacy. Transduction of avb3 integrin-expressing cell lines with wild type adenovirus Ad5CMVbgal showed that re-expression of avb3 integrin led to increased transduction efficacy. However, none of the transfectants were able to reach CA3ST, implicating that avb3 integrin up-regulation is not the only change responsible for increased adenoviral transduction efficacy. When transduction was made with adenoviral mutant Ad5FbD639, i.e. when the effect of CAR was partly eliminated, the transduction efficacy increased with avb3 integrin expression as well, but the maximal transduction efficacy was even higher than that of CA3ST cells, confirming that CAR up-regulation is also responsible for increased transduction of CA3ST cells. In conclusion our results suggest that increased adenoviral transduction efficacy in CA3ST cells is associated with increased expression of avb3.
