Sažetak
Mannose receptor (MR) is 175 kDa transmembrane receptor, a member of C type lectin receptor superfamily. With C-type lectin like domains it binds molecules ending with mannose, fucose or N-acetylglucosamine that are, in contrast to mammal cells, found on several microorganisms. With cistein rich domain MR binds sulfated sugars including N-acetylgalactosamine, chondroitin sulfate A and B and has been implicated in the clearance of hormones. MR facilitates both endocytosis and fagocytosis and is involved in antigen presentation. There are very few data about expression and function of MR at the maternal-fetal interface. The aim was to determine presence and endocytic activity of MR on antigen presenting cells in the human first trimester pregnancy decidua. Human first trimester pregnancy decidua tissues were obtained by elective pregnancy terminations. Tissues were either snap frozen for immunohistology studies or enzimatic digested followed by density centrifugation to obtain single mononuclear cell suspensions, which were used for cytospin slides and for flow cytometry. Endocytic activity of MR was measured by uptake of FITC-dextran. Using these methods we examined presence, distribution and endocytic activity of MR on decidual CD14 and CD83 positive cells. MR+ cells in the first trimester decidua were found mainly around decidual glands and blood vessels, and there is about 20% of MR+ cells in the cell suspensions of first trimester decidual tissue. About 74% of MR + cells are CD14+ macrophages, and approximately 12% are CD83+ dendritic cells. MR expression was found on 53% of decidual macrophages and on 12% of dendritic cells. Both, macrophages and dendritic cells in first trimester decidua were able to uptake FITC-dextran. Antigen presenting cells in the human first trimester decidua express MR and are able to mediate endocytosis in MR dependent manner.
