Sažetak
Miscanthus Anderss. is one of the most investigated genera for biomass production in Europe with the aim of reducing the CO2 loaded in the atmosphere. One of the most important goals in Miscanthus breeding is the improvement of combustion-related traits such as mineral content. Chlorine (Cl) and potassium (K) are considered to be of great importance in fouling, ash slagging and corrosion processes during biomass combustion. The objective of the research was to locate quantitative trait loci (QTLs) controlling Cl and K content in M. sinensis Anderss. with the aim of developing a Marker Assisted Selection (MAS) program. A population of 89 F1 hybrids from a cross between full-sibs P1 and P7 originating from a cross between MS-90-2 and MS-88-110 of M. sinensis was used for mapping. A linkage map based on 257 RAPD markers was developed using the 'offspring cross' mapping strategy (Atienza et al., 2002). The population was evaluated in a field trial at Córdoba, Spain following a randomized block with two replications in two consecutive years. Each field plot consisted of three clonally propagated plants. The mineral content was determined as a percentage of Cl and K in dry matter of plant stems. QTL analyses were performed using MapQTL 4.0 package (Van Ooijen et al., 2000) that allows the detection of QTLs in cross-pollinating populations. In a cross-pollinating population (i.e. Q1Q2xQ3Q4) up to four different QTL alleles can segregate. Thus, for each putative QTL three deviance effects were calculated, P1 and P7 effects (difference in effect of the alleles inherited from P1 and P7, respectively) and interaction effect (deviation from additivity), as described by Sewell et al. (2000). Six QTLs were detected for Cl and K considering both years. Out of these, four (Cl1, Cl2, Cl3, and Cl4) were detected for Cl while only two (K1 and K2) were detected for K. Cl1 was detected on the same chromosome region in both years, but the LOD peaks did not exactly coincide in both years. The rest of QTLs for chlorine, Cl2, Cl3, and Cl4, were only detected in a single year. K1 and K2 were only detected in year 2000, while no significant QTL was detected for K in 2001. Co-localization of QTLs for Cl and K was not detected although the phenotypic values were significantly correlated in both years. All the QTLs showed non-zero interaction values that would suggest strong dominance and/or epistatic effects. Cl1 was found over both years of analysis and therefore, it may be considered as stable. The rest of QTLs, expressing in a single year, could be dependent on environmental conditions and/or could be age dependent. As the mineral content is thought to be dependent of plant age in Mischantus, the first year may be considered as the establishment period while the second would correspond to a fully established crop. In conclusion, this study should be considered as a preliminary work to evaluate the possibilities to develop a MAS program for improvement of combustion-related traits in Mischantus.
