Sažetak
Spontaneous mutants of S. rimosus were characterised which showed changes in the 387 kb linear plasmid pPZG101 (Gravius et al., 1994). In one case an end of pPZG101 had integrated into the linear chromosome which resulted in the replacement of one chromosome end with a plasmid end. A second mutant carried a 1 Mb plasmid-prime in which one end of the linear plasmid had been replaced by a chromosome end. This resulted in a plasmid (pPZG103) carrying the oxytetracycline synthesis cluster which lies 600 kb from the chromosome end. In S. lividans 66 mutants were observed that had lost one or both chromosome ends. Mutants that had lost both chromosome ends had also lost the linear plasmid SLP2 (Lin et al., 1993), which has one end identical to the chromosome ends. SLP2 could not be reintroduced into such mutants. This suggests the existence of chromosomal genes in the deleted regions that are needed for stable replication of linear ends.
