Sažetak
We have established three embryogenic lines of pumpkin (Cucurbita pepo L.): a. auxin independent line PEDC - induced and maintained on hormone free MS medium with only reduced nitrogen source ; b. auxin dependent line DEC - induced and maintained on 2, 4-D containing MSC medium ; and c. habituated line HEC - induced on MSC medium with 2, 4-D, than transferred and maintained on hormone-free medium. The proces of embryogenesis of those lines coulde be control, except by auxin, by modification of reduced/unreduced nitrogen. We have analyzed the changes of cytidine methylation in three embryogenic lines of pumpkin cultivated on medium with or without auxin (2, 4-D and IAA) or with modified nitrogen sources. The results presented showed that two different control factors, reduced nitrogen (NH4Cl) and 2, 4-D affected the embryogenesis and provoked high level of DNA methylatrion. The high methylation was not only a consequence of the auxin treatment but a consequence of process which is related to the embryogenesis.
