Sažetak
Serotonergic mechanisms play an important role in the pathogenesis of migraine. Platelet serotonergic elements demonstrate pronounced similarities with the analogous structures in serotonergic neurons and therefore blood platelets might reflect serotonergic changes as a neuronal level. Platelet serotonin (5HT) level, kinetics of membrane 5HT transporter and kinetics of platelet MAO-B activity were measured in migraine patients with (n = 15) and without (n = 28) aura during the headache-free period as well as during the attack and compared to sex and age matched controls (n = 81). This work was based on our previous studies of platelet parameters in a large human population. Platelet 5HT level in migraine patients with aura (mean  SD): 654  123, without aura: 609  163, controls: 585  154 (ng 5HT/109 platelets) ; platelet 5HT uptake (Vmax) in migraine with aura: 145  17, without aura: 140  29, controls: 101  16 (pmol 5HT/108 plt./min), (Km) in migraine patients with aura: 0.393  0.080, without aura: 0.392  0.006, controls: 0.360  0.061 M) ; and platelet MAO-B activity in migraine patients with aura: 17.28  4.80, without aura: 16.01  5.2, controls: 16.32  6.7 (mol kynuramin/108 plt/60 min) during the headache-free periods. Our results clearly show the absence of any significant difference in serotonergic parameters in platelets of migraine patients during headache-free periods as compared to controls. On the other hand, an indication of potential changes in some of the mentioned parameters during migraine attacks was noted but conclusions cannot be made so far, because of too small numbers of patients studied.
