Sažetak
The aim of the study was to compare the expression of CD95 on peripheral blood (PB) CD4+ and CD8+ T-lymphocytes between patients with relapsing-remitting multiple sclerosis (RRMS) in the relapse of disease and healthy controls. The study included 8 patients (5F, 3M) with RRMS diagnosed by McDonald's criteria. Mean age of the patients was 34.88± ; 4.1 years, mean duration of the disease 3.5± ; 4.97 years, and mean EDSS scale 3.0± ; 0.8. Control group consisted of 10 healthy subjects (6F, 4M) mean age 33.7± ; 8.81 years. Quantitative assessment of PB T-lymphocytes bearing CD95 was done by use of monoclonal antibodies (anti-CD4-PE/Cy5, anti-CD8-FITC and anti-CD95-PE, DAKO) and flow cytometry (FACSCan cytometer and CellQuest software, Becton Dickinson). On statistical analysis nonparametric Mann-Whitney test for independent samples was used. Results were presented as median and range. A p value <0.05 was considered statistically significant. The proportion of CD8+ T-lymphocytes was significantly higher in patients in the relapse of the disease as compared with controls (M=37.65% ; range 33.3%-48.5% vs. M=29.75% ; range 21.1%-35.2%, p=0.003). The CD4/CD8 ratio was significantly lower in patients in the disease relapse as compared with controls (M=1.08 ; range 0.64-1.44 vs. M=1.56 ; range 1.13-2.65, p=0.01). The proportion of CD8+CD95+ T-lymphocytes was significantly higher in patients in the disease relapse than in controls (M=31.6% ; range 25.4%- 39.1% vs. M=22.55% ; range 12.3%-31.3%, p=0.01). The proportion of CD4+ T-lymphocytes was not significantly different between patients in the disease relapse and controls. However, the proportion of CD4+CD95+ T-lymphocytes was significantly lower in patients in the disease relapse than in controls (M=40.5% ; range 30.5%-47.9% vs. M=48.2% ; range 37.3%-55%, p=0.05). Our preliminary results show different expression of CD95 molecule on peripheral blood CD4+ and CD8+ T-lymphocytes in patients with RRMS, implicating impairment in the process of apoptosis. Since there are contradictory data on this issue in the literature, further studies are required to elucidate the pattern of CD95 expression and apoptosis in patients with multiple sclerosis.
