Sažetak
Aujeszky's disease virus (ADV) causes a natural infection in swine. For veterinary use vaccine against ADV is produced by continuous cell line such as Baby Hamster Kidney (BHK-21) cells or Porcine Kidney (PK-1) cells. While these cells are serum dependent during the growth process serum is used to enhance cell growth. In order to reduce amount of added serum we investigated influence of protein growth factor (PGF) obtained in our Laboratory on cell growth kinetics. We found that addition of PGF reduces population doubling time from 14, 7 h^-1 for control cells to 11, 9 h^-1 for PGF treated cells. The viability of the cells remained unchanged about 95-98 % in both cases. Production of vaccine against Aujeszky disease (AD) included biomass production of BHK 21 C13 cells where the cells are host material. When cells reached expected concentration, suspension of gE^- ADV Bartha K/61 was added into culture medium, and then started to entry into the cell as well as biological process During the further cultivation viability of cells was examined and that was the parameter for detection of ADV multiplication. Metabolic parameters like glucose and glutamine consumption and formation of lactate and ammonia are also measured. At the end of production, supernatant was suitable for production of live vaccine while from the cell precipitate it was possible to producte of inactivated vaccine. In all samples virus titre was measured earlier and expressed as TCID_50/mL. Addition of PGF during the biomass production and ADV multiplication has shown increased cell resistance on the ADV influence what was resulted in higher TCID_50/mL.
