Sažetak
Gene-targeting is the most important molecular tool for genome manipulations. However, the efforts for reliable genetically modified genomes are undermined by many factors influencing the ratio between the allelic and the ectopic recombination that controls the targeting fidelity. The known fact that this ratio is usually low in non-Saccharomyces yeasts and, most importantly, metazoans renders the understanding of the targeting mechanisms one of the key goals of contemporary genetics. Therefore, in this study I compared earlier side results of molecular construction of yeast strains with the contemporary knowledge focusing on the orientation of the insertion within the cassette for two reasons: (i) researchers usually choose the insertion orientation at random, and (ii) the possible influence of the parameter has never before been analyzed systematically. Thus, here I show that the cassettes bearing divergently oriented insertions significantly remedied mistargeting caused by their convergent counterparts in two loci (URA3 and LEU2) and one allele (ura3-52) in wt yeast cells (Mitrikeski, 2022, Curr. Genet., in press). It is tempting to suggest that the effect might be transcription-related. If so, this could potentially open a new area of genetics.
