Sažetak
During bone injury and healing, oxidative stress is present with lipid peroxidation and consequently formation of 4-hydroxynonenal (HNE). HNE can be toxic, but also a growth-modulating factor changing gene expression, cell differentiation and proliferation. Insulin-like growth factor, IGF-1 is synthesized by osteoblasts and has important role in osteoblasts proliferation and differentiation as well as bone growth and development. The purpose of our study was to see influence of HNE as mediator of oxidative stress on IGF-1 synthesis in mesenchimal cells of different origin, comparing normal and malignant cell reaction. For this purpose we have used human osteosarcoma HOS cells, WI38 human fibroblast cells and primary culture of human osteoblasts. HNE had different effects on IGF-1 synthesis in cells of different origin, indicating differential sensitivity and processes in those cells. As IGF-1 is very important autocrine and paracrine growth regulator of mesenchimal cell we conclude that differential response to HNE was caused by different processes in cells of different origin. This differential response was probably connected to their primary function(s) and has influence on bone growth and development in vivo.
