Sažetak
Prostate cancer (PCa) represents a most diagnosed neoplasmamong men, with incidence rates steadily rising. There is a needfor more sophisticated biomarkers for diagnosis, prognosis andmanagement. MicroRNAs (miRNAs) in liquid biopsies and tis-sue emerged as potential biomarkers for PCa that could help bet-ter distinguish aggressive vs. non- aggressive tumors, andmalignant vs. non-malignant conditions. However, conflictingdata was reported for certain miRNAs and published papersoften lack certain information which is necessary to contextualizethe results. With growing body of evidence, questions are beingraised regarding diverse (pre)analytical factors influencingmiRNA analysis and hinder translation of miRNA into clinicalpractice. In order to analyze and address current problems inmiRNA clinical research on PCa, a PubMed-based literaturesearch was conducted with the last update in May 2019. Diverse(pre)analytical factors influencing miRNA analysis were studiedand compared across the studies. By examining published papers, we conclude that studies widely differ in study design parameters:control groups, serum and plasma comparison, sample storage, quality control steps, data normalization. There is an immenselack of data on methodology framing parameters used in pub-lished studies. There is a lack of consistency and reproducibility, largely due to a missing consensus on preferred sample collec-tion, sample handling, RNA extraction and miRNA analysis.Other aspects such as different control groups and sample stor-age also influence variable results obtained in miRNA research.Of outmost importance seems to be reaching a consensus onguidelines and widely accepted protocols for sample processingand quality control. After identifying critical variations in designsand protocols that undermined clear-cut evidence acquisition, wepropose specific guidelines for critical steps that should be con- sidered in future research of miRNA as biomarkers especially inPCa.
