Sažetak
Appearance of transient cortical lamination serves as a basis for initial arrangement of cells to assemble first levels of cortical organization. Cell aggregation is one of the key events in the establishment of transient cortical lamination, characterized by differential gene expression of diverse layer-enriched markers. In addition, formation of early cortical compartments depends on instructions from extracellular environment, as well as from cell intrinsic properties. Our objective was to study early neurogenetic events that occur during transition from embryonic to fetal development in the prospective human frontal lobe. Immunofluorescence (IF) analysis was done on FFPE sections of postmortem human brain tissue from 7.5 and 8 postconceptional weeks (PCW), as a part of Zagreb Neuroembryological Collection. Imaging was done by confocal microscope Olympus FV3000. Results show that already at 7.5 PCW cell aggregation leads to sublayer formation in the late preplate (PPL) stage. Two types of neuronal population in cortical anlage were detected: TBR1+ glutamatergic and GAD67+ GABAergic neurons. Initial CP is formed at 8 PCW and TBR1+/DCX+ neurons were detected migrating towards CP to reach their final position. Moreover, fibrilar marker SMI312 showed first axonal fibers stretching through the CP. In conclusion, studying early neurogenetic events such as cell aggregation and initial CP formation is essential for a better understanding of neuronal identity specification, as well as the establishment of functional cortical connectivity.
