Sažetak
Epstein-Barr virus (EBV) belongs to the Herpesviridae family. Its certain latency stages are characterised by the expression of an LMP-1 oncogene which protein imitates CD40 on B- lymphocytes. Six LMP-1 variants are defined by mutations in C-terminal region: China1, China2, North Carolina, Alaskan, Mediterranean with deletion and Mediterranean without deletion. The aim of this study was to analyse diversity of LMP- 1 polymorphisms in pediatric patients with acute state of EBV infection represented as infectious mononucleosis (IM). Peripheral blood samples, from 34 children with IM were used. C-terminal region was amplified by PCR and analysed with Sanger sequencing. Obtained sequences were compared with a reference sequence and analysed with bioinformatics alignment tools. In total 14/34 sequences were wild type, 6/34 North Carolina and 3/34 Mediterranean without deletion. 3/34 patients had coinfection with wild type and North Carolina while 7/34 had coinfection with wild type and China1. According to the Mann-Whithey U test, concentration of C-reactive protein was higher in coinfection of LMP-1 variants (median 13 mg/L) compared to the infection with a single variant (median 9, 25 mg/L, p=0, 048). The ratio of CD4+/CD8+ was higher in coinfection (0, 3) compared with single variant (0, 2, p=0, 05). The results showed LMP-1 diversity in IM and association between different LMP-1 variants and parameters of immunological response.
