Sažetak
The histochemical reactions, as the part of complex incestigations of the constitution of vegetative bud of Norway spruce, were done to prove the presence or absence of lignin, pectic substances, cellulose, callous, lipids, basic proteins and DNA in the speific parts of the bud. The distribution of lipids, basic proteins and DNA during winter, the early spring and spring phase of the vegetative bud development into the shoot will be discussed in this paper. Lipids were stained with auramone O, basic proteins with Ponceau 2R and DNA with Hoechst 33342, Hoechst 33258 and DAPI on the 3 um thick longitudinal sections of vegetative buds of Picea abies (L.) Karst. which were fixed in 6% glutaradehyde. Lipids were present in embryonic pith cells and in loose cells in the cup-like structure. There were almost no lipids in the crown cells. Proteins rich in basic amino-groups were situated in nuclei, in cell and nucleus membranes but less in cytoplasm and cell walls. Cells with big, round nuclei were present in embryonic shoot during the winter phase. The cells of the crown had almost no nuclei. The wall of the cup-like structure contained a very small number of cells with round nuclei but the inside cells hac slim nuclei on the side. In the spring phase three zones of cells in the embryonic shoot were prominent: dermal zone of small cells with round nuclei ; zone of larger vacuolated cells with slim nuclei in mid-cortex ; and some cells with big, round nuclei in the middle of leaf primordia.
