Sažetak
Background and Purpose:Majority of hormones acting in the proximal tubule bind to their receptors in basal-lateral plasma membrane (BLM). However, the existane of functionally operated receptors in brush-border plasma (BBM), such as ET_B receptors which activate phospholipase C (PLC), was also demonstrated. The present study was performed to determine a possible existence of cross-talk between signalling systems in BLM and BBM. Materials and Methods: Rat renal cortical BLM were isolated using Percoll gradient method, while BBM were isolated by the Mg^2^+/EGTA-precipitation method. Diacylglycerol (DAG) concentration in BLM and BBM was measured using purified DAG kinase. Results and Conclusion: Cross talk between signalling systems localized in renal BLM and BBM was demonstrated. Sarafotoxin S6c-(S6c) mediated heterologous regulation of hepatocyte growth factor (HGF) signalling in BLM may be mediated by protein kinase C (PKC). On the other hand, HGF-mediated heterologous regulation of S6c signalling in BBM may be mediated by phosphoinositide 3-kinase (PI3K) and/or by one of its downstream signalling events.
