Sažetak
The development of medicines is an elaborative task that demands a great deal of time and expertise in a broad range of physical and medicinal sciences. In this contribution, numerous aspects of drug development and production will be discussed. A particular focus will be laid on solid oral dosage forms. The development of generally drugs starts with the conceptual design of the target molecule, which is most commonly performed by medicinal chemists and biologists. Once the target molecule is identified, the research directions will go through a very complex labyrinth of synthetic and analytical evaluations before the molecule’s solid-state properties are considered in the crucial final steps of the research and development process. The understanding and full control over the solid- state properties of small molecule pharmaceutical agents (which excludes proteins and other macromolecules) plays are central role in the development of solid oral dosage forms. Small molecule drug candidates are known to exist in the form of various polymorphs, solvates or hydrates, as well as in amorphous phases. In addition, a broad range of phase transitions (e.g. polymorph interconversions, desolvation of solvates, etc.) can occur during pharmaceutical processesing, thus altering the desired physicochemical properties (e.g. dissolution rate, transport characteristics, morphology) of the drug candidate. Active pharmaceutical ingredients (APIs) are nowadays most conveniently developed and delivered as solid dosage forms. It is, therefore, not surprising that solid-state forms of APIs are regarded as immensely important not only form a clinical point of view, but also from legal and regulatory perspectives. The last 50 years witnessed extensive an systematic investigations in the area of pharmaceutical solid-state chemistry that led to the establishment of procedures for the systematic evaluation, understanding and control of solid-state properties of drugs. These investigations also resulted in the discovery of thousands of different crystalline forms of APIs, and thereby contributed to the wealth of knowledge and experience that presently allows the industry to craft efficient, safe and optimised drugs. This contribution also highlights the significant achievements in research and development of pharmaceutical solids in PLIVA in the last twenty years.
