Sažetak
The treatment with direct acting antivirals (DAA) for hepatitis C infection (HCV) has offered an opportunity to analyze kinetics profiles of serum biomarkers and the changes in the immune system. The role of complex biological response modulators in the immunopathogenesis of HCV have not been sufficiently investigated. The aim of this study was to analyze the serum biomarker levels prior and after DAA treatment in HCV chronic- infected patients. Serum concentrations of 13 growth factors (Angiopoietin-2, EGF, EPO, FGF- basic, G-CSF, GM- CSF, HGF, M-CSF, PDGF-AA, PDGF-BB, SCF, TGF- α, VEGF) and 12 cytokines (IL-5, IL-13, IL-2, IL-6, IL-9, IL-10, IFN-γ, TNF-α, IL-17A, IL- 17F, IL-4 i IL-22) were analysed in 56 chronic HCV patients before and after sustained virological response (SVR) and compared with 15 controls by using bead-based flow cytometry. Statistical analysis was performed using R. Statistical significance was set at p< ; 0.05. The results shown the importance of variables in the classification of patients before and after DAA treatment compared with controls. Significantly increased concentrations of IL-4, SCF, and IL-5 were observed in patients with chronic hepatitis C before and after DAA tretment. The association of SCF and IL-4 in patients with HCV infection compared to controls suggests an important role for mast cells in HCV. Also, IL-4, SCF and IL-5 could be considered as a specific biomarkers of DAA treatment. Therefore, differences in SCF concentrations between HCV patients and controls are extremely important, as SCF is one of the key factors in liver regeneration.
