Sažetak
The stable gastric pentadecapeptide BPC157 (GEPPPGKPADDALV, M.W. 1419) currently in clinical trials for inflammatory bowel disease (PLD-116, Pliva), also ameliorates internal and external wound healing. Now, it also accelerates Achilles tendon healing associated with functional, biomechanical, microscopical, and clinical recovery in rats. Likewise, in vitro, it stimulates growth of cultured rat tendocytes treated in vitro with supraphysiological amounts of 4-hydroxynonenal (HNE), an end product of lipid peroxidation and a mediator of oxidative stress. The right Achilles tendon was transected 5 mm proximal to its calcaneal insertion. Large tendon defect appears between cut ends due to muscles retraction. Assessments were made either daily (Achilles Functional Index (AFI)) or on day 4, 7, 10, 14 (biomechanical, microscopical and macroscopical). Medication (BPC 157 (dissolved in saline, with no carrier addition) (10 ľg, 10 ng or 10 pg/kg b.w.) or saline (5.0 ml/kg)), was given intraperitoneally, once daily, first 30 min after surgery, the last 24 h before autopsy. Controls generally have severely compromised healing. In rats treated with pentadecapeptide BPC 157 rats recovery is presented with (i) biomechanically, increased load of failure, load of failure per area and Young elasticity modul ; (ii) functionally, significantly higher AFI-values ; (iii) microscopically, advanced fibroblasts, reticulin and collagen formation ; (iv) macroscopically, smaller size and depth of tendon defect, and subsequently the reestablishment of full tendon integrity. Thus, in vivo accelerated tendon healing and in vitro stimulated growth of tendocytes, the simple way of application and peptide stability, may give gastric pentadecapeptide BPC 157 potential for routine application in future therapy.
