Sažetak
In the 4-year period (1998 &#8211 ; 2002) indigenous Croatian grapevine cultivars were examined for phytoplasmas. Both visual and molecular surveys were performed on cultivars from two coastal vine-growing regions, Istria and Dalmatia, where autochthon cultivars are prevalently grown. Samples representing 7 Vitis vinifera L. cultivars were included in this study: &#8216 ; Borgonja&#8217 ; , &#8216 ; Debit&#8217 ; , &#8216 ; Istarska Malvazija&#8217 ; , &#8216 ; Maraština&#8217 ; , &#8216 ; Plavina&#8217 ; , &#8216 ; Plavac mali&#8217 ; and &#8216 ; Teran&#8217 ; . Grapevines showing symptoms characteristic of grapevine phytoplasmoses were collected as well as the asymptomatic plants. Total nucleic acids were extracted from leaf midribs or cane phloem scrapings and used for molecular amplification of phytoplasma 16S rRNA gene. A general phytoplasma primer pair R16F1/R0 was used for direct PCR. The first nested molecular amplification was performed with another phytoplasma generic primer pair (R16F2/R2) which was then followed by another round of nested PCR employing either universal phytoplasma primers (16R738f/16R1232r) or primers R16(I)F1/R1 specific for the phytoplasma ribosomal groups 16SrI (Aster Yellows) and 16SrXII (Stolbur or Bois Noir). Amplification products from all nested PCR experiments were singly digested with restriction endonucleases MseI or Tru9I, AluI and KpnI. RFLP patterns were compared to those of standard phytoplasma strains. During the first two years of this survey phytoplasma associated disease symptoms were not observed in Istria and Dalmatia. Molecular analyses confirmed the absence of phytoplasmas from grapevines grown in these regions. However, in the years that followed, grapevine yellows-like symptoms did appear in Dalmatian cultivars &#8216 ; Debit&#8217 ; , &#8216 ; Maraština&#8217 ; and &#8216 ; Plavina&#8217 ; and in some of the Istrian cultivars. In a Dalmatian cultivar &#8216 ; Plavac mali&#8217 ; no phytoplasma-like symptoms have been observed so far. PCR and RFLP results for &#8216 ; Debit&#8217 ; were consistent with possible phytoplasmosis symptom observations. Phytoplasmas belonging to the ribosomal group 16SrXII (Stolbur or Bois Noir), subgroup XII-A have only been identified in samples of &#8216 ; Debit&#8217 ; , a Croatian indigenous cultivar. No phytoplasma ribosomal DNA has been amplified from asymptomatic samples of &#8216 ; Plavac mali&#8217 ; . Visual observations indicating the phytoplasma presence in all other cultivars included in this study still awaits the molecular confirmation.
