Sažetak
Process of inflammation resolving includes immune cell activation and recruitment at the site of the inflammation, the co-operation of lipid mediators, which originate from essential n-3 (anti- inflammatory) and n-6 (pro-inflammatory) PUFAs, as well as cytokine microenvironment present. Due to their involvement in the inflammatory response, the intake ratio between n-3 and n-6 PUFAs is of great importance for homeostasis. Lately, there has been a shift in Western diet towards increased intake of n-6 PUFAs and a decline in n-3 PUFAs intake which consequently changes the balance between pro- and anti-inflammatory markers. Forty young, healthy volunteers consumed three hard-boiled eggs per day and kept a diet diary for three weeks (N (regular eggs) = 21 ; N (n-3 PUFA eggs) = 19). Samples were taken on two separate occasions, before and after the respective protocols. Lipid mediators’ serum concentrations were measured with commercially available ELISA assays while cytokine concentrations were measured with antibody-based, magnetic bead reagent kits on Luminex platform from serum samples and supernatants from PMA-Ionomycin-treated PBMC cell cultures. RvE1 endpoint serum concentration significantly increased (p=0.013), while prostaglandin E2/E3 serum concentrations ratio significantly decreased (p=0.014) in the n-3 PUFA group compared to the baseline. INF-γ significantly decreased and IL-10 significantly increased in serum samples of the n- 3 PUFA group. Furthermore, TGF-β1 production by PBMCs significantly increased (p=0.026) following n-3 PUFAs dietary protocol, while IL-6 production was significantly decreased (p=0.05), compared to the respective baseline levels. Diet supplemented with n-3 PUFAs alters immune response towards inflammation resolving conditions through effects on lipid mediators and cytokine secretion following a significantly changed serum fatty acid composition and n-6/n-3 PUFAs ratio.
