From year to year, the demand for natural and environmentally friendly cosmetics is growing rapidly, and thus the interest in herbal extracts with biologically active ingredients that can prevent and delay skin degradation. Considering fact that skin ageing is caused by the degrading effect of free radicals and oxidative stress, antioxidants such as flavonoids are important class of phytoconstituents. Jasione montana L. (Campanulaceae) is a rich source of polyphenolic compounds, main flavonoids such as luteolin, luteolin 7-O-glucoside and luteolin 7- O- sambubioside. Our previous reports confirmed the anti-tumour activity of J. montana. Taking these aspects into account, the antioxidant potential (DPPH assay) and enzyme inhibition activities that negatively affect the skin structure (elastase, collagenase) of glycerol extracts was investigated. Extracts with a variable amount of glycerol: 50% glycerol (JM50G), 60% glycerol (JM60G) and 80% glycerol (JM80G) were assessed in an in vitro model by spectrophotometric method. All tested extracts showed significant antioxidant properties during the DPPH assay. However, JM60G the most potent extract with an EC 50 value of 1.44±0.11 μL extract/mL was observed. Butylated hydroxyanisole (BHA) was used as a positive control with EC 50 = 3.26±0.06 μg/mL. However, two of the three tested extracts (JM50G and JM60G) showed significant activity against elastase with EC50 values of 64.01±1.91 and 52.54±2.45 μg/mL, respectively. The EC 50 for ursolic acid used as a positive control was 24.09±0.8 μg/mL. All tested extracts (JM50G, JM60G and JM80G) were effective for collagenase to a similar degree (EC 50 = 26.84±1.86, 23.88±1.01, 23.32±2.14 μg/mL respectively) and gallic acid (positive control) with an EC50 value of 124.70±9.59 μg/mL. Our results show that various extracts can be a promising component of natural preparations with anti-ageing properties that prevent and delay skin degradation.
