Sažetak
Mycotoxin ochratoxin A (OTA) is nephrotoxic in all experimental animals. The mechanism of OTA toxicity includes the effect on phenylanine metabolism and on the mitochondrial function. The other mode of action of OTA in kidney is oxidative damage by enhancing free radicals generation followed by increased peroxidation of lipids. The end product of lipid peroxidation is malondialdehyde (MDA). The nephrotoxic effect of OTA was investigated in adult female Wistar rats treated intraperitoneally with OTA (0.5 mg/kg b.w.). The animals were treated 3 times a week, and sacrificed twenty four hours after 1, 3, and 6 treatments, and 21 day after 12 treatments, respectively. The animals were kept in metabolic cages, and urine was collected during 24h period after 1st, 3rd, 6th, 9th, and 12th treatment and on day 21. after the 12th treatment. In kidney tissue the concentration of OTA increased gradually up to the 6th treatment, while the MDA concentration was the highest after the first treatment (219&plusmn ; ; ; 15 nmol/g tissue, Mean&plusmn ; ; ; SD), decreased gradually after 3rd and 6th treatment (188&plusmn ; ; ; 17, and 178&plusmn ; ; ; 16 nmol/g, respectively) and 21 day after 12th treatment the concentration of MDA did not differ from controls (152&plusmn ; ; ; 10 nmol/g tissue). MDA and OTA concentration in urine as well as the activity of enzymes leucine amino-peptidase, lactate dehydrogenase, isocitrate dehydrogenase and alkaline phosphatase increased until the sixth treatment, and returned to control values 21 day after the 12th treatment. Contrary to the MDA concentration in kidney, MDA in urine is the consequence of lipid peroxidation in the whole organism. Therefore, measurement of MDA in kidney, but not in urine, is good parameter of OTA-induced kidney lesion.
