Sažetak
Astrocytes, the most abundant and diverse neuroglial cells in the central nervous system, are powerful regulators of the brain environment. It has already been established that astrocytes influence synaptic plasticity as well as reorganize neural circuits after traumatic brain injury (TBI). The aim of this study was to test the effects of chemically functionalized single- walled carbon nanotubes (SWCNTs) on the expression of excitatory amino acid transporter 1 (EAAT1) and glial fibrillary acidic protein (GFAP) following in vitro rapid stretch injury of the primary mouse astrocytes. For the in vitro TBI, astrocytes were grown on silastic membranes and subjected to severe stretch. One hour following the injury, solutes containing SWCNTs or vehicle were added to the cell growth media. At 24 h post-TBI, cells were collected and lysates prepared for the western blot analyses. Our preliminary results suggest that the application of SWCNTSs to the severely injured astrocytes increased the protein expression of EAAT1, but had no significant effect on the GFAP levels. Our findings point to potential protective effect of SWCNTs following in vitro TBI, mediated through the increase in the EAAT1 expression.
