Sažetak
Decreased lower esophageal sphincter pressure and transient relaxations of the lower esophageal sphincter are common pathogenic factors in gastroesophageal reflux. Previously, pentadecapeptide BPC-157 (currently in clinical phase II for inflammatory bowel disease, PL-10, PLD-116, PL-14736, Pliva d.d.) has protective effect on esophageal mucosa in an animal model of GERD. Therefore, this study examines the effect of gastric pentadecapeptide BPC 157 on the lower esophageal sphincter and pyloric sphincter pressure. The study was done on 48 adult female Wistar rats. All animals were randomly assigned into 6 groups, 4 experimental (10 μ g/kg and 10 ng/kg pentadecapeptide BPC-157 i.g.) and 2 control (0.9% saline). All rats underwent an operation to implant a Foley catheter into the stomach, 3 groups through esophageal and 3 groups through duodenal incision. Proximal side of the esophageal or distal side of the duodenal incision was ligated to prevent the regurgitation of the stomach content. Water manometer, connected to the drainage port of the Foley catheter, measured the positive pressure of the pentadecapeptide BPC-157 or saline, applied directly into the stomach, increasing the intragastrical pressure. Manometrical evaluation showed that pentadecapeptide BPC 157, in both concentration, given through the catheter under the positive pressure, into the stomach significantly increased the basal lower esophageal sphincter pressure (p < 0.01) and it significantly decreased the basal pyloric sphincter pressure (p < 0.05) as compared to control. The results suggest that the gastric pentadecapeptide BPC-157 may decrease reflux episodes of reflux esophagitis by increasing basal lower esophageal sphincter pressure and by accelerating gastric emptying.
