Sažetak
Introduction: Fatty acid derived lipid mediators have an important role in maintaining cytokine environment through direct effects on cytokine production and T lymphocytes. In the present study, we aimed to determine the effects of n-3 PUFA supplementation (~1053 mg/per day) through consumption of enriched hen eggs, on pro- and anti-inflammatory markers in healthy individuals (N=40 ; 23.8 ± 2.57 years old). Materials and Methods: Participants consumed three regular (N=21) or enriched hen eggs (N=19) per day through three weeks. Mononuclear cells (PBMCs) were isolated from whole blood by density gradient centrifugation and cultured in RPMI-1640 media (10% FBS ; 1% antibiotic). Concentrations of anti-/pro-inflammatory cytokines (IL-10, IL-6, IL- 23, IL-17A, and TGF-β1) and MCP-1 chemokine in both control and n-3 PUFA group, before and after dietary protocols were determined in supernatants following PMA/ionomycin stimulation with Invitrogen ProcartaPlex antibody-based, magnetic bead reagent kits on the Luminex 200 instrument platform. Results: TGF-β1 production by PBMCs following n-3 PUFA dietary protocol was significantly increased (p=0.026), while IL-6 production was significantly decreased (p=0.05), compared to the respective baseline levels. Target cytokine and chemokine production was unaffected by the consumption of regular hen eggs. Conclusion: The results of the present study have demonstrated that dietary n-3 PUFAs can modify key markers of inflammation and lead towards inflammation resolving environment in the population of young, healthy individuals.
