Sažetak
Ophiostoma novo-ulmi is a phytopathogenic fungus that is causing Dutch elm disease (DED), a devastating vascular wilt disease of elms. This aggressive pathogen has been spreading in a form of two subspecies: O. novo-ulmi ssp. novo-ulmi and O. novo-ulmi ssp. americana. These subspecies show only weak prezygotic barrier to hybridization and are expected to readily hybridize in nature, resulting in the emergence and spread of ssp. novo-ulmi x ssp. americana hybrids in overlap zones. In order to investigate population structure of DED pathogen in Croatia, O. novo-ulmi isolates infecting host species Ulmus laevis, U. minor and U. glabra from five locations were characterised utilising PCR-RFLP analysis and sequencing of the cu and col1 genes. Analysed cu and col1 gene sequences had 2 and 6 variable sites, respectively, which allowed us to differentiate the two subspecies, but the variability of these genes within subspecies has not been found. Molecular identification of O. novo-ulmi isolates revealed presence of both subspecies in Croatia with significantly higher incidence of ssp. novo-ulmi compared to ssp. americana. Presence of hybrids was confirmed at all investigated sites, with 27% of total number of tested isolates being identified as hybrids. The virulence of O. novo- ulmi isolates was assessed using rapid virulence assay by inoculation of apple (Malus x domestica ‘Golden Delicious’) fruits. Significant variation in the size of the necrotic lesion induced by tested fungal isolates indicates that Croatian O. novo-ulmi isolates exhibit variable virulence level what could have significant implications on DED epidemiology.
