Sažetak
Mammalian embryos cultivated in vitro develop in teratomas during two weeks. We developed a modified organ culture system in our laboratory which provides favorable conditions for the study of differentiation of embryonic cells in response to various exogenous factors that could prevent or direct the differentiation and growth. W e tested impact of retinoic acid (RA) and dibutyryl cAMP (db-cAMP) on differentiation of cultivating mouse embryo in critical stage of development when it is extremely sensitive to various extrinsic factors. Retinoic acid was added in various concentrations to the culture medium. The period of embryos exposition to RA was 98 hours and 9 days. The other group of embryos was exposed 48 hours to RA and after that to db-cAMP until the end of culture. RA inhibited chondrogenesis and changed the differentiation pattern of epithelia. Combined treatment with RA and db-cAMP increased myotube differentiation. Therefore, in embryo-derived teratomas in vitro retinoic acid and db-cAMP altered gene expression, shown by change in the differentiation pattern of various tissues.
