Sažetak
THE USE OF MERISTEM TIP CULTURE TO ELIMINATE VIRUSES FROM STRAWBERRY PLANTS- We tested a technique for virus elimination by meristem tip culture. With this method we have succeeded in producing healthy plants. We took very small pieces (0, 2 - 0, 3 mm) of apical meristem and we planted them in M.S. nutrient medium (Muraschige and Skoog, 1962) suplemented with organic constituents of glucose and agar (Boxus, 1974). It was a basic medium. On this medium suplemented with 1 mg/l IBA, 0, 1 mg/l 6-BAP we succeeded in getting sprouts in 40-50% of cases. The sprouts were aseptically transfered into a basic medium with the adition of IBA 1 mg/l and 6 - BAP 1 mg/l. After 4-6 weeks of incubation in a climate chamber, the sprouts were multiplied by forming axillary buds so there were 10-15 new sprouts. The sprouts from axillary buds were transfered into basic medium with the addition of 1 mg/l IBA where they produced roots and leaves. After next 4-6 weeks 95% of the plants formed roots. Those plants were tested for viruses. It was confirmed that no economicaly major virus diseases were present. Until recently strawberries were not grown on large areas in Yugoslavia, although the enviromental conditions are satisfactory. During the last few years a greater interest has aroused for this culture. The production of it is insufficient for our contry's needs. In order to expand production some new strawberry farms are necessary which require healthy young strawberry plants. We tried to produce them from existing cultivars. For our experiments we used cultivars Senga, Sengana and Gorella which came from productive farms. These plants were used as meristem donators. The whole process "in vitro" was divided into three phases: a) Meristem culture b) Sprouts multiplication c) Rooting These three phases were followed by the acclimatization of new plants and their testing on viruses. The meristem medium which was used for a)b)c) mentioned above consisted of inorganic salts (Murashige and Skoog, 1962), organic constituents of 40 g/l glucose and 6 g/l agar.
