Sažetak
Helicobacter pylori is a Gram-negative microaerophilic bacterium that chronically colonizes the gastric epithelium. This pathogen is responsible for the development of several gastrointestinal diseases ; including non-symptomatic chronic gastritis, peptic ulcer disease, gastric mucosa-associated lymphoid tissue (MALT) lymphoma, and gastric adenocarcinoma. Adenylosuccinate synthetase (AdSS) is one of the essential enzymes in purine salvage pathway. It is a ligase which catalyzes the condensation of IMP with L-aspartate to form adenylosuccinate by hydrolyzing GTP in the presence of Mg 2+ ions. Since Helicobacter pylori lacks the genes for de novo synthesis of purines, as shown in bioinformatic studies, AdSS is a potential drug target. The aim of the project is biochemical and structural characterization of this enzyme and to find potential new inhibitors, which could be efficient in eradication of this pathogen.
