Sažetak
Presented results are the part of continuous investigations of spruce vegetative buds composition and changes during their proliferation in spring shoots, in order to better understand their physiological responses in tissue culture, e.g. the dependence of successful adventitious buds induction and the period of taking the initial explant. The concentration of photosynthetic pigments was determined in isolated vegetative buds or needles of Picea abies and Picea pungens. Chlorophylls a, b and carotenoids were extracted in 80% acetone and measured by spectrophotometer on = 452.5, 645 and 663 nm. Sampling was done weekly during the year from summer to spring. results showed the uniform level of chlorophyll a+b concentration in the vegetative buds during the summer, autumn and winter (P. abies: 0.15-0.35 mg/g fresh weight, P. pungens: 0.15-0.25 mg/g), what was 10 times lower than in needles developing from those buds in spring (P. abies: 1-2 mg/g fresh weight, P. pungens: 1-1.5 mg/g). After increasing time, end of spring, chlorophyll concentration in needles was continuous during the year. The ratio of chlorophyll a/b in buds was 1-1.5, and in needles was 2-2.5. Concentration of carotenoids was higher in needles (0.4-0.5 mg/g), than in buds (about 0.1 mg/g). Established differences between P. abies and P. pungens might be caused by different genotype or they can be specific for a species, what should be further topic.
