Sažetak
Using laser ablation laser ablation, we show that in U2OS cells the kinetochores can segregate without the attachment to spindle pole or any other microtubule structure of the spindle. We found that major contribution to kinetochore movement in isolated fibres came from “pacman” depolymerisation and sliding between bridging fibres connecting sister k-fibres, while contribution of poleward flux was negligible. Our results demonstrate that the bridging fibre, while linked to the k-fibres, can contribute to chromosome separation by pushing apart sister kinetochores through sliding mechanism.
