Sažetak
Glycans are complex, highly branched carbohydrate structures composed of monosaccharide sugars. They are typically found either attached to proteins (glycoproteins) or lipids (glycolipids). Glycosylation, the process of covalent bonding of glycans to a protein, is a common co- and post-translational modification. Most biopharmaceuticals are glycoproteins and include monoclonal antibodies, erythropoietin, cytokines, etc. Control of glycosylation is of major importance during the development of these drugs, because their glycan chains have marked effects on stability, activity, antigenicity, and pharmacodynamics. The glycan structure-function relationship of many glycoproteins is still not fully understood. Therefore, it is imperative to investigate their functional importance, so that once glycoengineering tools are available at affordable costs, targeted remodeling of protein glycosylation can become an integral part of manufacturing the next-generation of biopharmaceuticals.
