Sažetak
Natural killer (NK) cells dominate among synovial tissue lymphocytes of patients undergoing primary total knee replacement surgery. We investigated synovial tissue distribution of NK cells in regards to CD 14+ cells and activation status of NK cells in regards to cytotoxic mediators in patient with osteoarthritis. Mature synovial tissue was sampled from patients undergoing primary total knee replacement surgery. Tissue was embedded in paraffin cubes and cut in 5-6 um thick tissue sections. Single labelling of CD56, NKp46, CD14, HLA-DR, CD16 and CD 161, as well as double labeling of CD56 and cytotoxic mediators, perforin, granulysin, TNF-related apoptosis induced ligand (TRAIL) and Fas Ligand (FasL) were performed by immunofluorescence respectively. Distributions of NK cell markers CD56 and NKp46 were similar to each other. NK cells are on the surface, sublining synovial tissue and accumulate around the CD 14+ cells in the stroma of synovial tissue shoots. HLA- DR+ and CD 16+ cells were found in the accumulations of CD 14+ macrophages and NK cells. Almost all CD56+ cells express granulysin and perforin, while TRAIL and particularly FasL have been rarely expressed. CD161 expressing cells are on the surface of synovial tissue. Synovial NK cells of patients with osteoarthritis seem to be activated (CD161+) in the close vicinity of cognate macrophages and well equipped with cytolytic (perforin) and apoptotic (granulysin) mediators.
