Endocytosis of MHC class I molecules is not yet well defined process. There is evidence indicating that the mechanisms of spontaneous and induced internalization are not the same. Mechanism of internalization of MHC class I molecules induced by viral proteins differs from the spontaneous internalization. This may represent a useful model for further studies of the endocytic process. The aim of our study was to compare the kinetics of spontaneous and induced internalization and intracellular fate of different H2-d molecules. Spontaneous endocytosis of Ld molecules in P815 cell line, treated with cycloheximide continuously through the experiment, and by MCMV induced endocytosis of Kd and Dd in mouse embryonic fibroblasts were monitored by flow cytometry and western blotting analysis. Half time of Ld molecules on the cell surface is 6 hrs, and after 24 hrs most of them are internalized. Biochemical analysis after 24 hrs revealed almost the same quantity of fully conformed Ld molecules as in the beginning, while the quantity of non-conformed (empty) Ld declined at the same period of time. Kd and Dd molecules showed similar kinetics as fully conformed Ld molecules, while those infected by MCMV showed tendency to be degraded. Spontaneous internalization of H2-d molecules is not followed by degradation, differently of those internalized by MCMV.
