Sažetak
Under conditions of phosphate starvation the PHO5 promoter of S. cervisiae undergoes a striking chromatin transition, whereby a chromatin microdomain consisting of four positioned nucleosomes is completely disrupet ( Svaren and Hörz, 1997). How this remodeling event is achieved is not yet understood. It is assumed that Pho4, the main transcriptional activator of PHO5, recruits transcriptional coactivators to the promoter including ATP-dependent remodeling machines as well as histone acetyltransferases. Although Gcn5 is not required for full activation of PHO5 the rate of PHO5 activation is greatly decreased in its absence (Barbaric et al., 2001). This delay in transcriptional activation is specifically due to slow chromatin remodeling of the PHO5 promoter, whereas the transmission of the phosphate starvation signal to the PHO5 promoter progresses at a normal rate. A role in affecting the rate of chromatin remodeling seems to be a more general property of chromatin modifying activities because a similar delay in PHO5 activation is observed when Snf2 is absent. Again the final extent of activation and chromatin remodeling is very similar to the level seen in a wt strain. We have asked the question, which cofactors are recruited at what time to the PHO5 promoter upon inductoin. Chromatin immunoprecipitation (ChlP) analysis employing tagged versions of the Pho4, Ada2 and Snf2 proteins was used to determine the kinetics with which these factors bind to the promoter. Data on how the recruitment of one coactivator affects the recruitment ot others is presented. The contribution and significance of SAGA and of histone acetylation for chromatin remodeling at the PHO5 promoter is discussed.
