Sažetak
World supplies of fossil fuels are decreasing and, due to the environmental pollution caused by their excessive use, it is necessary to find a more economical and a more sustainable alternative. Fermentation of renewable lignocellulose substrates to ethanol by the yeast Saccharomyces cerevisiae is one of the alternatives for fuel production. Since yeast cannot directly ferment lignocellulose substrates, it is first necessary to hydrolase lignocellulose in order to release fermentable sugars. Moreover, during the pretreatment of lignocellulose biomass, different inhibitors that slow down or inhibit growth of microorganisms are formed. Therefore, various attempts have been made to construct a production strain that will be resistant to those growth inhibitors. In this research, the yeast strain Žup ura- was transformed with the pSP-YAP1-C620F plasmid that enables overexpression of constitutively active gene YAP1C602F that, for some strains of the yeast S. cerevisiae, increases resistance to growth inhibitors. Constructed strain with overexpression of YAP1C620F allele indeed displayed increased resistance to acetic acid and 2-furaldehyde in comparison to a strain that has overexpression of the wild type YAP1 gene.
