When studying optimalconditions for the in vitro inhibition of human monocyte spreading (a test for cell-mediated immunity), we determined several features of the spreading. Monocytes from human peripheral blood spread equally in medium 199 and in the same medium containing two per cent bovine serum albumin or horse serum globulin. The soreading ofmonocytes from 84 persons was distribzted normally after three hr incubation in medium 199, with 60 - 69 per cent being the most frequent range. Spreading percentages in individual chambres varied less if the values were above 65. Age (64 persons from a few months to 75 years) had no influence on the monocyte spreading. onocytes of months to 75 years had no influence on the monocyte spreading. Monocytes of 38 male and 34 females spread is similarly, as did monocytes of 50 sick persons(acute and chronic glomerulonephritis, rheumatoid fecer, melano, a, carcinoma) and 27 healthy persons ofvthe same age and sex.
