Sažetak
The aim of this paper is to investigate if variations in the histological method of processing samples could lead to better students’ recognition, interpretation and understanding. Students watch and draw 90 microscopic slides at the histological practicals so at the end of the course they should be able to recognize and define different microscopic structures of tissues and organs. Three samples of lymph nodes from two pigs were taken and processed by selected histological methods at the Faculty of Veterinary Medicine, University of Zagreb. Two samples of lymph nodes were taken from the pig A. They underwent fixation, dehydration and embedding in paraffin by hand. After that, they were separated into two units. The first unit was trimmed and sectioned with a microtome SLEE MAINZ CUT 5062 and stained according to hematoxylin and eosin method by hand. In the end, there were eight microscope slides and the best one was chosen to be Sample no. 1. Second unit was trimmed and sectioned with a microtome microTec cut 400 and stained according to H&E method by Microm HMS 70. We choose the representative sample out of four slides that we had got and named it Sample no. 2. Another sample was taken directly from the slaughterhouse from the pig B. It was immersed in liquid nitrogen and cut using a cryostat Thermo Shandon. The sections were placed on glass slides and stained according to H&E method by hand. We got eleven microscope slides and we choose the best one to be Sample no. 3. Five demonstrators and two assistants analyzed three samples and filled the table. The table consists of eight parameters which include: a typical structure, capsule, trabecula, cortex, medulla, reticular cells, lymphatic nodules and lymphocytes. They could give 0-3 points to each parameter due to visibility and staining. Although Sample no. 2 achieved the highest score, there were no significant differences in the quality of samples due to variations in histological methods.
